He made the announcement in front of supporters at Structural Steel. The Republican has been serving in the Secretary of State's office since 2008.
Hosemann says he wants to provide a better future for his grandchildren in Mississippi.
"The lieutenant governor has a unique position in state government, to control the budget and all the statutory duties," said Hosemann. "It's a very powerful position, and it'll leave me an opportunity to make a real difference in Mississippi."
No other Republicans have qualified to run against Hosemann. State Representative Jay Hughes is the only Democrat to qualify so far for the position.
Thursday, Hughes called on Hosemann to resign from his current role to preserve the integrity of the election.
